Antimicrobial and antibiofilm efficacy of tetrasodium-EDTA complexes against several common wound pathogens.

Abstract: There is increasing evidence showing the presence of biofilms on surfaces, such as medical devices and wounds, can lead to infection, inflammation and delayed wound healing. Development of chronic wounds following infection can significantly increase patient morbidity and mortality as well as the economic burden on healthcare, as biofilms often show increased tolerance to antimicrobials and antibiotics in comparison to their planktonic counterparts and are therefore difficult to treat. The aim of this study was to evaluate the antibiofilm efficacy of Tetrasodium-EDTA complexes against several common wound pathogens. The minimum inhibitory concentration (MIC) of T-EDTA alone and in several different complexes with silver and various metal ions was determined using a broth microdilution method. The minimum biofilm eradication concentration (MBEC) was determined following ASTM E2799-17 Standard disinfectant efficacy against Pseudomonas aeruginosa biofilm using the MBEC assay. Synergistic interactions between components of the complexes was evaluated using a 2D checkerboard MIC method and determining the fractional inhibitory concentration index (FICI).Following MIC and MBEC testing of T-EDTA alone and in complexes with silver and various metal ions, increased antimicrobial and antibiofilm potency was found with all complexes tested. Checkerboard assays showed synergistic interactions between several silver compounds and T-EDTA, with an FICI of 0.2 – 0.5 against Pseudomonas aeruginosa and Staphylococcus aureus. The data presented here demonstrates potent antibiofilm activity of T-EDTA complexes against several common wound pathogens in planktonic and biofilm states. The efficacy against biofilms should be investigated further, for potential application to treating and preventing infection in healthcare settings.
